Single-Cell RNA Sequencing Reveals Human iPS Cell-derived Alveolar Type 1 Cells in Alveolar Organoids
Updated January 23, 2023We induced differentiation from human induced pluripotent stem cell (hiPSC)-derived alveolar type2 (AT2) cells into hiPSC-derived 2D alveoalr type 1 (2D-iAT1) cells. Single cell RNA sequencing revealed that 2D-iAT1 cells were composed of three clusters. Overall design: After isolation of hiPSC-derived AT2 cells, hiPSC-dervied AT2 cells were cultured in two-demensional condition for 7 days, and then single cell libraries were generated using Fluidigm C1 system. Single "2D_EpCAM+" cells were captured and separated on the each chamber of medium-sized (17-25 µm) C1 integrated fluidic circuit (IFC) with the C1 system (Fluidgm) Then, first-strand cDNA was synthesized and amplified on the independent chamber using SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing.
